The aim of this study was to investigate its antiangiogenesis activity and its inhibitory effect on the apoptosis of human umbilical vein endothelial cells (HUVECs) induced by tumor necrosis factor-related apoptosis-inducing ligand (TRAIL). We used Western blot and real-time PCR to determine whether HA14-1 inhibited Bcl-2, p53, and XIAP expression, which regulate TRAIL-induced apoptosis in HUVECs. The inhibitory activity of HA14-1 on angiogenesis in vitro was investigated by matrigel-based angiogenesis assay in HUVECs. The cytotoxicity of HA14-1 to HUVECs was determined by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay. HA14-1 did not cause significant cytotoxicity in HUVECs. The expression of Bcl-2, p53, and XIAP was inhibited by HA14-1 in a time- and dose-dependent manner. HA14-1 treatment inhibited TRAIL-induced apoptosis and decreased the proliferation of HUVECs. More importantly, HA14-1 had a stronger antiangiogenesis activity than a p53 inhibitor, PFT-α, and an XIAP inhibitor, HA14-1-Glo.
